De collectie omvat inmiddels zo’n 7000 voorwerpen waarvan er 400 in het museum te zien zijn. De overige getuigenissen van het culturele erfgoed van Brielle en omgeving worden bewaard in de museumdepots.De collectie is geregistreerd in het collectieregistratiesysteem Adlib. De komende jaren wordt gewerkt aan aanvulling en actualisering van de gegevens.
Het voornemen is de gehele collectie via internet te ontsluiten; binnen korte tijd zal de deelverzameling kunstvoorwerpen via internet toegankelijk zijn.
Het Historisch Museum Den Briel beoogt nog steeds via schenkingen, bruiklenen en aankopen voorwerpen te verzamelen die een materiële getuigenis vormen van de geschiedenis van Brielle en omgeving. Dit kunnen prenten, tekeningen of schilderijen zijn maar ook driedimensionale voorwerpen.
Een van de belangrijkste aanwinsten voor het museum, mede dankzij de steun van velen waaronder de Vereniging Rembrandt, was in 2002 de aankoop van het portret op paneel van de Brielse jurist Cornelis Briel, in 1643 geschilderd door David Bailly (1584-1657). Cornelis Briel bekleedde verschillende bestuursfuncties, zoals schepen van Brielle, baljuw van Brielle en Voorne, en tevens was hij curator van de Latijnse school in Brielle.
In 2012 bestaat het museum honderd jaar. Het museum zal die verjaardag vieren met de realisatie van een uitbreiding van de depots en een aansprekend heringericht museum. Daarnaast zal de publicatie Honderd Jaar Verzamelen worden uitgegeven.
In de museumwinkel zijn artikelen te koop die verband houden met de museumcollectie en/of de geschiedenis van Brielle: ansichtkaarten, boeken, dvd’s, speurtochten etc.
